Title: ANTICANCER AND APOPTOSIS INDUCTION PROPERTIES OF APIUM GRAVEOLENS SEEDS

Abstract:

Based on traditional medicinal use Apium graveolens seed was selected to evaluate its anti-cancer property with special reference to apoptosis induction, if any. When the water, alcohol and hexane extracts of the dried powder of the plant seeds were tested for cytotoxicity to Dalton’s Lymphoma Acitic (DLA) cells in vitro using Tryphan blue method, only the n-hexane extract showed significant activity at 500 µg/ml level. The extract induced apoptosis as evidenced by morphological changes. The cytotoxicity of this extract was found to be more to cancer cells (DLA) compared to normal cells (thymocytes and macrophages). An active chloroform fraction was separated from this extract. The active fraction at a dose of 200 mg/kg protected 75 % of the mice challenged with 1 X 106 DLA cells whereas all untreated control mice died of cancer. Thin Layer Chromatography (TLC) on silica gel further separated this anticancer fraction into 2 cytotoxic components. One of them was identified as a steroid positive component whereas the other was an alkaloid positive component. The steroid component induced apoptotic cell death while the other component induced necrotic cell death.  These components are attractive materials for further studies leading to possible anti-cancer drug development.

    1. have you check only n-hexane effect on DLA cell ?

    2. the effect of various compounds on DLA is one thing and its side effects on nervous tissue is another issue ? isnt it .......so one should select such plant products which we use routinly.........

    It is really great to learn that we have some potential anti cancer medicinal plants in our traditional medicine system. We must encourage to go for further research so that cheaper medicines are available for already financially overburdened cancer patients.

    We checked with 3 extracts of water, alcohol and n-hexane. The water extract did not show any toxicity to DLA cells up to 500 μg/ml. However, the hexane extract showed 48% cell death at 50 μg/ml whereas alcohol extract showed almost the same level of toxicity at 500 μg/ml. The cell death was associated with nuclear condensation, membrane blebbing and formation of apoptotic bodies (characteristics of apoptotic cell death).

    Yes, Dr. Ajit V Pandya. That’s true. While selecting the drugs, we tried our best to choose the medicinal plants which are cheap and routinely available. In our primarily study, we identified the apoptotic induction effect of Apium Gravelones seed (commonly known as Ajmud in Hindi) in n-hexane extract. Although it induced apoptosis, the apoptotic mechanism remains to be studied. Detailed studies in various experimental cancer models as well side effects on nervous tissue are required to establish its efficacy.

    Thank you very much for your review. Based on the literature review, we have selected 4 plants for screening to identify apoptosis inducing properties. They are Abrus Precatorius Linn, Acorus Calamus Linn, Apium Graveolens Linn, Piper Longum Linn. Further, the cytotoxicity effect of all above plants extracts of water, alcohol, n-hexane were tested on Thymocytes, Macrophages and DLA cells and identified that n-hexane extract of Apium graveolens having the properties of cytotoxicity effect and membrane blebbing activity.

    Thank you very much for your review and comment. We haven't come through any articles for nano-level properties of Apium Graveolens seeds. Just for the curiosity, I have googled and found that one article where leaf extract of Apium Graveolens were used for biosynthesis of silver nanoparticles from silver salt (interesting and please find below link).
